| 體外活性 | 在節(jié)細(xì)胞層,Mdivi-1具有強(qiáng)烈的免疫反應(yīng)性.在缺血誘導(dǎo)12小時(shí)后的GCL中,Mdivi-1可使蛋白表達(dá)增加.Mdivi-1使GFAP蛋白表達(dá)顯著降低,并但不改變Drp1蛋白表達(dá).在正常的小鼠視網(wǎng)膜中,Mdivi-1主要分布在內(nèi)網(wǎng)層、節(jié)細(xì)胞層、外網(wǎng)層及內(nèi)核層.在缺血小鼠視網(wǎng)膜中的早期神經(jīng)發(fā)育中,Mdivi-1和膠質(zhì)纖維酸性蛋白的蛋白表達(dá)會(huì)明顯增加.Mdivi-1可阻斷缺血視網(wǎng)膜中的細(xì)胞凋亡,并使缺血兩周后的RGC存活率顯著增加. |
| 體內(nèi)活性 | Mdivi-1通過變構(gòu)調(diào)節(jié)阻斷了Dnm1的ATP酶活性和自組裝（IC50<10 μM）。在HeLa細(xì)胞和細(xì)胞外鼠源肝細(xì)胞線粒體配置液中,Mdivi-1有效地抑制了C8-Bid和STS誘導(dǎo)的MOMP（線粒體外膜通透性增加）。Mdivi-1抑制酵母Dnm1和人源Drp1分裂DRPs(發(fā)動(dòng)蛋白相關(guān)GTP酶),且高效可逆地誘導(dǎo)線粒體融合進(jìn)一個(gè)巢樣的結(jié)構(gòu)。在細(xì)胞中,Mdivi-1通過抑制線粒體外膜通透性從而抑制細(xì)胞凋亡。Mdivi-1大體上代表了一類**中風(fēng)、心肌梗死和神經(jīng)退行性疾病的療法。 |
| 激酶實(shí)驗(yàn) | All GTPase assay reactions are started in a 200 μL volume, of which 150 μL is placed into the well of a 96-well plate. Depletion of NADH, as monitored by reading the A340 of the reaction, is measured every 20 s for a total of 40 min using a SpectraMAX 250 96-well plate reader. Spectrophotometric data are transferred to Excel and the measured steady state depletion of NADH over time is converted to protein activity. |
| 細(xì)胞實(shí)驗(yàn) | Mdivi-1 is dissolved in DMSO. YPGlycerol plates are topped with 10 mL YPGlycerol containing 1% low melt agar and 75 μM mdivi-1, and cells are spotted 12 hours later using a 48 well pinning device. After pinning cells, plates are incubated at 24°C or 37°C and imaged using an Eagle Eye II imaging system. |
