| 產(chǎn)品描述 | Ketanserin is a quinazoline derivative and serotonin (5-hydroxytryptamine, 5HT) receptor subtype 2 (5-HTR2) antagonist with potential antihypertensive and antiplatelet activities. Following administration, ketanserin binds to and inhibits the signaling mediated by 5-HTR2, which inhibits serotonin-dependent vasoconstriction and platelet activation. |

| 體內(nèi)活性 | 研究發(fā)現(xiàn)在外側(cè)膝狀體核中,Ketanserin可使α1 - 腎上腺素能受體介導(dǎo)的反應(yīng)以及去甲腎上腺素引起的興奮性反應(yīng)程度降低,其是誘導(dǎo)而非減弱5-HT的抑制作用。在分離的大鼠尾動脈,頸動脈,犬基底,胃脾動脈和冠狀動脈,犬隱靜脈和胃脾靜脈中,Ketanserin可劑量依賴性抑制5-羥色胺誘導(dǎo)的收縮反應(yīng)。在大鼠心肌細胞中,Ketanserin使動作電位時程顯著增加,復(fù)極可從50％達218％、90％達256％,且對其它動作電位參數(shù)無顯著影響。Ketanserin抑制連接后α-腎上腺素能受體激活犬隱靜脈和大鼠尾動脈的收縮。Ketanserin抑制灌注豚鼠胃血管對5-羥色胺的收縮反應(yīng),在某些實驗中還可逆轉(zhuǎn)該反應(yīng)。Ketanserin以時間和濃度依賴性方式抑制Ito電荷區(qū)（EC50：8.3 μM）。Ketanserin還以劑量依賴性方式抑制Ito和持續(xù)電流（EC50：11.2 μM）,但不影響L型鈣電流和內(nèi)向整流鉀電流。 |
| 細胞實驗 | Ketanserin is dissolved in DMSO to produce a stock solution of 100 mM. Ketanserin stock is diluted in experimental solutions to achieve the final concentrations[1]. The established HEK 293 cell line stably expressing hERG channels is cultured in Dulbecco's modified Eagle's medium (DMEM) supplemented with 10% foetal bovine serum, 400 μg/mL G418. The HEK 293 cell line stably expressing recombinant human cardiac KCNQ1/KCNE1 channel current (IKs) is maintained in DMEM containing 10% foetal bovine serum and 100 μg/mL hygromycin. Cells used for electrophysiology are seeded on a glass coverslip. The mutant hERG channels are constructed, and are transiently expressed in HEK 293 cells using 10 μL of Lipofectamine 2000 with 4 μg of hERG mutant cDNA in pCDNA3 vector[1]. |
