| 體外活性 | 10058-F4在脂肪、肺、肝臟和腎臟中含量高.10058-F4的峰值濃度至少比血漿峰值濃度低10倍.10058-F4的分布容積>200 ml/kg,終端半衰期約為1 h.10058-F4（20/30 mg/kg,i.v.）對(duì)小鼠生長(zhǎng)無(wú)明顯抑制作用.10058-F4（i.v.）在5 min時(shí)達(dá)到血漿峰值（300 μM）,在6 h時(shí)濃度低于檢測(cè)下限. |
| 體內(nèi)活性 | 10058-F4使AML細(xì)胞停滯在G0/G1期,使c-Myc的表達(dá)下調(diào)、CDK抑制劑p21和p27上調(diào)。同時(shí),其通過(guò)激活線粒體途徑誘導(dǎo)細(xì)胞凋亡,出現(xiàn)Bcl-2下調(diào),Bax上調(diào),胞漿內(nèi)細(xì)胞色素C釋放,及caspase3/7/9裂解。此外,10058-F4可能通過(guò)激活多種轉(zhuǎn)錄因子從而誘導(dǎo)骨髓細(xì)胞分化。相似地,10058-F4可誘導(dǎo)原代AML細(xì)胞凋亡和分化。10058-F4使c-Myc蛋白水平降低并抑制HepG2細(xì)胞增殖,可能與上調(diào)細(xì)胞周期蛋白依賴(lài)性激酶抑制劑p21WAF1及降低細(xì)胞內(nèi)[α]-甲胎蛋白的水平有關(guān)。10058-F4還可在轉(zhuǎn)錄水平使人端粒酶逆轉(zhuǎn)錄酶下調(diào)。除抑制HepG2細(xì)胞增殖外,10058-F4還增強(qiáng)其對(duì)常規(guī)化療藥物,阿霉素, 5-氟尿嘧啶和順鉑的敏感性。 |
| 細(xì)胞實(shí)驗(yàn) | Cells, plated in 96-well plates (105/mL for cell lines and 5 × 105/mL for primary leukemic cells), are treated in triplicate with indicated concentrations of 10058-F4. At various time points, 20 μL 5 mg/mL MTT is added to each well. After incubation at 37°C for 3 hours, the MTT medium is removed and 100 μL DMSO lysis buffer is added. The number of viable cells is assessed by the percentage of absorbance of treated cells relative to that of solvent controls, using 570-nm wavelength on a spectrophotometer.(Only for Reference) |
